Diversified DNA Libraries for Antibody, Peptide and Nanobody library construction, Methods&Technique

Generating libraries of diversified oligonucleotides, especially those with sequences containing randomized codon regions, is a critical step in the development of antibody, peptide, and nanobody libraries. These libraries are foundational tools in therapeutic discovery, enabling the identification of molecules with high specificity and affinity for a wide range of targets. The methods or technologies used for generating these libraries have evolved to allow high levels of diversity, specificity, and efficiency. The currently available methods and techniques for generating such libraries are: Solid-Phase DNA Synthesis Trimer Phosphoramidite-Based Synthesis & Considerations Regarding Trimer Phosphoramidite Technology PCR-based Random Mutagenesis DNA Shuffling Microchip-based DNA Synthesis Oligonucleotide Ligation CRISPR-Cas9 Mediated Library Generation Cell-Free Transcription-Translation Systems Phage Display with Randomized Peptide Libraries All these will be discussed in detail in this podcast.
